03Mechanism of action
CJC-1295 DAC
CJC-1295 is a GHRH analogue carrying a drug affinity complex (DAC) that binds albumin after injection, extending its half-life to days. It stimulates pituitary GHRH receptors and raises GH and IGF-1.
Ipamorelin
Ipamorelin is a synthetic pentapeptide that releases growth hormone through the ghrelin (GH secretagogue) receptor, like GHRP-6. It was developed from a GHRP-1 series with the central Ala-Trp dipeptide removed.
04Study endpoints
CJC-1295 DAC
- Two randomized placebo-controlled trials in healthy adults: a single injection raised mean GH 2- to 10-fold for 6 days or more and IGF-1 1.5- to 3-fold for 9–11 days.
- Estimated half-life was 5.8–8.1 days; after repeated doses IGF-1 stayed above baseline for up to 28 days.
- GH pulsatility was preserved: trough GH rose 7.5-fold, mean GH rose 46% and IGF-1 rose 45%, with pulse frequency and size unchanged.
Ipamorelin
- In rat pituitary cells and in anaesthetised rats and conscious swine, GH-release potency and efficacy were similar to GHRP-6.
- In swine it did not change FSH, LH, prolactin or TSH, and unlike GHRP-6 and GHRP-2 it did not raise ACTH or cortisol across a wide dose range.
- In adult female rats, 15 days of treatment dose-dependently increased longitudinal bone growth (42 to up to 52 µm/day) without changing total IGF-I.
