The short version

Both are investigational. Survodutide (GLP-1 + glucagon) reported up to 14.9% mean weight reduction at 46 weeks and liver benefits in a MASH trial. Retatrutide (adds GIP) reported up to 24.2% at 48 weeks. These were separate trials with different designs, so neither result proves one is stronger.

01

How they work

Survodutide activates GLP-1 and glucagon receptors. Retatrutide activates those two plus the GIP receptor. Glucagon signalling is thought to raise energy use and liver fat burning; adding a receptor does not by itself prove a bigger clinical effect.

02

What studies measured

Survodutide's obesity trial (387 enrolled, 386 treated) reported −6.2% to −14.9% mean weight change across active groups versus −2.8% with placebo at 46 weeks; only 233 of 386 completed treatment.

Its MASH trial (293 treated, biopsy-confirmed fibrosis F1–F3) found MASH improvement without fibrosis worsening in 47%, 62% and 43% of active groups versus 14% with placebo at 48 weeks.

Retatrutide's obesity trial (338 adults) reported −7.2% to −17.5% versus −1.6% at 24 weeks (primary endpoint) and −8.7% to −24.2% versus −2.1% at 48 weeks.

03

Reported side effects

Gastrointestinal events dominated both programs. In survodutide's obesity trial they affected 75% versus 42% on placebo; in the MASH trial nausea was 66% versus 23% and vomiting 41% versus 4%.

Retatrutide events were mostly mild-to-moderate gastrointestinal effects, with dose-related heart-rate increases.

04

Evidence limits

There is no head-to-head trial. Different populations, durations, dropout rates and analysis methods mean the headline percentages cannot be ranked against each other. Phase 3 results are needed for both.
