01

Mechanism of action

Cagrilintide is a long-acting analogue of amylin, a pancreatic hormone that induces satiety. It is studied alone and combined with semaglutide (CagriSema).

02

Observed research endpoints

  • Phase 2 (26 weeks): every cagrilintide arm reduced weight more than placebo (6.0%–10.8% vs 3.0%); the top arm also beat liraglutide (10.8% vs 9.0%).
  • REDEFINE 1 (3,417 adults without diabetes): cagrilintide–semaglutide gave −20.4% mean weight change at 68 weeks vs −3.0% with placebo.
  • REDEFINE 2 (type 2 diabetes): −13.7% vs −3.4% with placebo at 68 weeks.

03

Regulatory & safety context

  • Investigational: not approved as a standalone medicine.
  • Phase 2: gastrointestinal events in 41%–63% vs 32% on placebo, mainly nausea. REDEFINE 1: gastrointestinal events in 79.6% vs 39.9%.
