01

Mechanism of action

SS-31 (elamipretide) is a mitochondria-targeted tetrapeptide that binds cardiolipin in the inner mitochondrial membrane and is studied for stabilising mitochondrial energy production.

02

Observed research endpoints

  • MMPOWER-3 (218 adults with primary mitochondrial myopathy, 24 weeks): no improvement vs placebo in 6-minute walk distance or fatigue.
  • An earlier crossover trial missed its primary endpoint but showed signals in patient-reported outcomes, which supported MMPOWER-3.

03

Regulatory & safety context

  • Not approved for mitochondrial myopathy; research-grade material is not a medicine.
  • Generally well tolerated in trials; injection-site reactions were the most common event (80% in the crossover trial), mostly mild.
