01

Mechanism of action

Tesofensine is a small-molecule (not peptide) triple monoamine reuptake inhibitor (dopamine, noradrenaline and serotonin), originally studied for Parkinson's and Alzheimer's disease.

02

Observed research endpoints

  • Phase 2 obesity trial (24 weeks): mean weight loss of 4.5%–10.6% across arms vs 2.0% with diet and placebo.
  • A mechanistic RCT found strong appetite-suppressing effects, a 4.6% higher night-time energy expenditure and greater 24-hour fat oxidation vs placebo.

03

Regulatory & safety context

  • Not approved by the FDA or EMA for any indication; research-grade material is not a medicine.
  • Monoamine reuptake inhibition is associated with heart-rate and blood-pressure increases, which were monitored in trials.
