Cagrilintide is a long-acting analogue of amylin, a pancreatic hormone that induces satiety. It is studied alone and combined with semaglutide (CagriSema).

• Phase 2 (26 weeks): every cagrilintide arm reduced weight more than placebo (6.0%–10.8% vs 3.0%); the top arm also beat liraglutide (10.8% vs 9.0%).
• Combination evidence, not cagrilintide-alone efficacy: REDEFINE 1 (3,417 adults without diabetes) reported −20.4% mean weight change with cagrilintide–semaglutide vs −3.0% with placebo at 68 weeks using the treatment-policy estimand.
• Combination evidence in type 2 diabetes: REDEFINE 2 reported −13.7% mean weight change with cagrilintide–semaglutide vs −3.4% with placebo at 68 weeks, not a standalone cagrilintide result.

• Investigational: not approved as a standalone medicine.
• Phase 2: gastrointestinal events in 41%–63% vs 32% on placebo, mainly nausea. REDEFINE 1 combination arm: gastrointestinal events in 79.6% vs 39.9% with placebo.
