03Mechanism of action

Tesofensine is a small-molecule (not peptide) triple monoamine reuptake inhibitor (dopamine, noradrenaline and serotonin), originally studied for Parkinson's and Alzheimer's disease.

04Study endpoints

  • The 24-week phase 2 obesity trial reported mean weight loss 4.5–10.6 percentage points greater than diet and placebo across active arms; diet and placebo alone produced 2.0% mean loss. The publication carries a 2013 expression of concern.
  • A mechanistic RCT found strong appetite-suppressing effects, a 4.6% higher night-time energy expenditure and greater 24-hour fat oxidation vs placebo.

05Safety & regulatory findings

  • Not approved by the FDA or EMA for any indication; research-grade material is not a medicine.
  • The 2008 obesity publication carries a 2013 Lancet expression of concern; its efficacy and safety findings require that qualification. Heart-rate and blood-pressure effects were monitored in trials.
