01

Mechanism of action

Oxytocin is a hypothalamic nonapeptide acting on oxytocin receptors in the uterus, breast and brain. Intranasal oxytocin is studied for social cognition, appetite and stress.

02

Observed research endpoints

  • Meta-analysis (466 participants with neurodevelopmental disorders): no significant effect on emotion recognition and a small significant effect on theory of mind.
  • Meta-analysis of appetite trials: single-dose intranasal oxytocin reduced food intake in non-psychiatric participants but not in anorexia, bulimia, binge-eating disorder or schizophrenia.

03

Regulatory & safety context

  • FDA-approved by injection (e.g. Pitocin) for obstetric uses; intranasal use for behaviour or appetite is not approved.
  • Reviewers call for larger, well-designed randomized trials.
